چکیده

The concurrent administration of Griffonia simplicifolia lectin into the peritoneal cavity of mice has been reported to protect them from challenge with Ehrlich ascites tumor cells. This study demonstrates that the GS I lectin is capable of binding to the surface of both inflammatory macrophages and Ehrlich ascites tumor cells and can mediate macrophage lectin-dependent cytotoxicity to this tumor cell line. The lectin was not found to be toxic to Ehrlich ascites tumor cells directly or to nonspecifically initiate complement-mediated lysis of these target cells. The macrophage lectin-dependent cytotoxicity could be specifically inhibited by the haptenic sugar methyl a-D-galactopyranoside. Thus, this lectin appears to be able to provide a linkage between effector macrophages and Ehrlich ascites tumor cells resulting in the triggering of a cytolytic event.

Inhibition of Ehrlich ascites tumor cell growth by Griffonia simplicifolia I lectin in vivo.

Griffonia (Bandeiraea) simplicifolia I (GS I) seeds contain a family of alpha-D-galactopyranosyl-binding isolectins which strongly agglutinate Ehrlich ascites tumor cells due to the presence of this determinant sugar on their cell surface glycoproteins. Administration of GS I lectin (100 micrograms/day or 300 micrograms on alternate days, i.p.) inhibits the growth of Ehrlich tumor cells in vivo...

Lectin and Neuropeptide Labeling in Whole-Mount Preparation of Meninges in the Rat

495 Abstract: The distribution of both peptidergic and non-peptidergic primary afferents in meninges was investigated in whole-mount preparations from meninges. Lectin Griffonia simplicifolia I-B4 (GSA I-B4) was used as a marker for non-peptidergic fibres. Calcitonin gene-related peptide (CGRP) was used as a marker for peptidergic fibers. Lectin Griffonia simplicifolia selectively labeled thin ...

Comparison of binding proteins on the surface of murine tumor cells for two lectins active in the lectin-dependent macrophage-mediated cytotoxic reaction.

The binding proteins for Sarcophaga lectin and wheat germ agglutinin on the surface of Ehrlich ascites tumor cells were compared. Studies with antibody against the binding protein for Sarcophaga lectin showed that these two binding proteins are different. Since these two lectins are both active in the lectin-dependent macrophage-mediated cytotoxic reaction with Ehrlich ascites tumor cells as ta...

An lnsecticidal N-Acetylglucosamine-Specific Lectin Gene from Griffonia simp/icifo/ia (Leguminosae)'

Griffonia simplicifolia 11, an N-acetylglucosamine-specific legume lectin, has insecticidal activity when fed to the cowpea weevil, Callosobruchus maculatus (F.). A cDNA clone encoding G. simplicifolia II was isolated from a leaf cDNA library, sequenced, and expressed in a bacterial expression system. The recombinant protein exhibited Kacetylglucosamine-binding and insecticidal activity against...
